The antigen gliadin is immobilized on the microtiter plate. During the first incubation step, the human anti-gliadin-sIgA antibodies in the samples are bound by the immobilized antigen. After a washing step, the anti-gliadin-sIgA antibody presence is determined by addition of a peroxidaselabeled anti-sIgA antibody. Tetramethylbenzidine is used as a peroxidase substrate. The enzymatic reaction is stopped by an acidic stop solution. The absorbance of the color compound is determined photometrically at 450nm. The measured absorbance is directly proportional to the amount of bound anti-gliadin-sIgA antibodies. The results are evaluated by comparison with a cut-off value.
