Retinol binding protein (RBP) is a low molecular weight whose primary function is the transport of vitamin A (retinol) from the liver to the periphery. The majority of RBP is bound to transthyretin, but a small circulating fraction is passed freely through the glomerulus in the kidney and metabolized by the proximal tubules. In the normal kidney, excreted RBP levels are very low, therefore, increased urinary excretion of the protein is indicative of kidney impairment.

The Mouse Retinol Binding Protein 4 ELISA (Mouse RBP4 ELISA) is a highly sensitive two-site enzyme linked immunoassay for measuring RBP4 in biological samples of mice.

In the Mouse Retinol Binding Protein 4 ELISA (Mouse RBP4 ELISA), the RBP4 present in samples reacts with the anti-KIM-1 antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-RBP4 antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound RBP4. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3’,5,5’-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of RBP4 in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of RBP4 in the test sample. The quantity of RBP4 in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.
