Biomarkers of Bone Metabolism
Bone metabolism is a constant battle of bone growth and bone resorption. These two events are carefully orchestrated by several key proteins.
Canonical Wnt-signalling plays an important role in the regulation of bone homeostasis by promoting the development of osteoblasts. Negative regulators of the Wnt pathway are important new therapeutic targets for the treatment of diseases with enhanced bone resorption. One of these molecules is Sclerostin (SOST), a 22.5 kD secreted glycoprotein, which acts by binding to the Wnt-coreceptor LRP5, thus preventing the binding of Wnt molecules. Sclerostin is almost exclusively produced in osteocytes and is therefore considered a highly specific clinical marker for bone. It has stimulated considerable interest as a potential therapeutic target in the study of diseases associated with low bone mass, including osteoporosis.

The Sclerostin ELISA was debuted in an abstract presented at the ASBMR meeting in 2009. The Sclerostin ELISA is the first commercially available SOST ELISA.
